Neurite Outgrowth Quantification Assay
Proper development and function of the nervous system requires differentiated neurons to extend neurite projections that give rise to dendrites and axons. Using an elegant system of molecular guidance cues and extra cellular matrix (ECM) proteins, these cellular outgrowths navigate long distances to form an intricate network of connections between each of over a trillion neurons present in the human brain. Understanding the mechanisms that mediate neuritogenesis is important for all aspects of nervous system function including proper development and pathologies associated with adult brain and spinal cord injury. Using microporous filter technology, Millipore provides a quick and efficient system for quantitative analysis of compounds that influence neurite formation and repulsion. The microporous filter allows for the biochemical separation and purification of neurites and cell bodies for detailed molecular analysis of RNA/ protein expression, signal transduction processes, and identification of drug targets that regulate neurite growth or retraction processes. The versatile assay design uses sensitive protein staining technology and permits highly quantitative analysis by optical density (O.D.) using a standard microplate reader. It is also possible to directly visualize neurite structures using fluorescent microscopy for specific protein changes, expression, and location.
