UniProt Summary
FUNCTION: SwissProt: Q15389 # Binds and activates TIE2 receptor by inducing its tyrosine phosphorylation. Implicated in endothelial developmental processes later and distinct from that of VEGF. Appears to play a crucial role in mediating reciprocal interactions between the endothelium and surrounding matrix and mesenchyme. Mediates blood vessel maturation/stability. It may play an important role in the heart early development.

Entrez Gene Summary
Angiopoietins are proteins with important roles in vascular development and angiogenesis. All angiopoietins bind with similar affinity to an endothelial cell-specific tyrosine-protein kinase receptor. The protein encoded by this gene is a secreted glycoprotein that activates the receptor by inducing its tyrosine phosphorylation. It plays a critical role in mediating reciprocal interactions between the endothelium and surrounding matrix and mesenchyme. The protein also contributes to blood vessel

Description:
Anti-Angiopoietin-1, N-terminus
Background Information:
The remodelling of the primitive vasculature in mammals to form the mature cardiovascular system includes a number of factors directing endothelial cell development. Angiopoietin-1 (mouse and human Ang-1; 498aa with 98% identity) is an angiogenic secreted protein that interacts with the endothelial specific Tie-2 receptor. It is primarily expressed in developing endothelial cells. During embryonic dvevelopment, Ang-1 binds and induces tyrosine phosphorylation of Tie-2. Ang-1 appears to play a crucial role in mediating matrix and mesenchyme. It mediates blood vessel maturation and stability. Ang-1 may play a critical role in heart development. Ang-1 deficient mice mimic the phenotype exhibited by animals deficient in Tie-2. A homolog of Ang-1, termed Angiopoietin-2 (mouse and human Ang-2, 496aa with 85% identity) may act as an antagonist for Ang-1 and Tie-2. Ang-1 and Ang-2 have ~60% sequence homology.
Alternate Names:
Ang-1
Specificity:
Recognizes mouse Angiopoietin-1 (Ang-1). The immunogenic peptide sequence is 90% (18/20 aa) conserved in human Ang-1, 95% I pig and bovine, and has no significant sequence homology with Ang-2. Antibody crossreactivity in other species is not established.
Key Applications:
- ELISA
- Western Blotting
Application Notes:
Western blot: 1-10 ug/mL using ECL. Expected size of Ang-1 is ~55kDa.
ELISA: 1:10,000-1:100,000 using 50-100 ng of control peptide per well (Catalog Number AG635).
Immunohistochemistry: Not tested. It is recommended that the antibody be used at 2-20 ug/ml in formaldehyde fixed tissue.
Optimal working dilutions must be determined by end user.
