Aquaporins are a family of small integral membrane proteins related to the major intrinsic protein (MIP or AQP0). This gene encodes an aquaporin which functions as a molecular water channel protein. It is a homotetramer with 6 bilayer spanning domains and N-glycosylation sites. The protein physically resembles channel proteins and is abundant in erythrocytes and renal tubes. The gene encoding this aquaporin is a possible candidate for disorders involving imbalance in ocular fluid movement.

Background Information:
Aquaporin 1 is a 28 kDa integral membrane protein which was originally identified in red blood cells and renal proximal tubules. AQP1 is also expressed by the choroid plexus and various other tissues. It forms a water-specific channel that provides the plasma membranes of red cells and kidney proximal tubules with high permeability to water, thereby permitting water to move in the direction of an osmotic gradient.

Application Notes:
Immunohistochemistry: 2-10 µg/mL on paraformaldehyde fixed tissue sections.

Western Blotting: 1-10 µg/mL using Chemiluminescence technique. Immunoblots reveal two bands, a lower 28 kDa band corresponding to unglycosylated form and an upper high molecular 35-60 kDa glycosylated component (Denker et al. 1988; Smith & Agre 1991) We recommend the use of 0.5-1.0% milk in all primary/secondary antibody-enzyme conjugate incubations in order to suppress non-specific bands.

ELISA: 0.5-1.0 µg/mL using 1 µg/mL control peptide (Catalog Number AG307)/well.

Optimal working dilutions must be determined by end user.

A 19 aa synthetic peptide from the cytosolic carboxyl terminal domain of the AQP1 (Lanahan et al. 1992; Deen et al. 1992; Moon et al. 1993; Patil et al. 1994) conjugated to KLH. This peptide contains most of the epitopes recognized by polyclonal antibodies against the whole protein (Smith & Agre 1991). The immunogen peptide is absolutely conserved in AQP1 from human, ovine, bovine, rat, and mouse.

This peptide is available for blocking experiments (catalog number: AG307).
