Description:
Anti-cyclic AMP (cAMP)
Specificity:
Specific for cAMP with no cross reaction to other nucleotides at concentrations to 1 mM. Reactive with cAMP fixed in tissues or acetylated cAMP. Not reactive with free, non-acetylated, cAMP in RIA or EIA assays.

Application Notes:
Immunohistochemistry: 1:1000 - 1:10,000, depending upon the detection method employed.

Radioimmunossay: Dilution of the antisera (50 µL) 1:3,000 will provide enough material to assay ~6000 tubes. Sensitivity is < 0.6 pg/50 µL in the acetylated cAMP RIA and the concentration of cyclic AMP producing 50% displacement from total binding is 1.5 ± .06 pg. Recommended protocol available upon request.

ELISA: 1:5,000 - 1:10,000. Acetylated cAMP or acetylated samples are required. Samples are acetylated as follows: In rapid succession, add 20:1 of triethylamine, followed by 10:1 acetic anhydride to each sample. Vortex. The procedure is performed at room temperature and may be done up to several hours prior to actually setting up the assay. Samples are diluted 50:1 with PBS prior to performing any assay.

Cyclic AMP (cAMP, adenosine 3´, 5´-cyclic monophosphate) is a nucleotide which acts as a key second messenger in multiple signal transduction pathways. cAMP regulates many cellular functions, such as metabolism, cell growth and differentiation, gene transcription, ion transport and ion channel function. It is synthesized from ATP by the action of adenylate cyclase, and is inactivated by hydrolysis to 5´-AMP by the actions of phosphodiesterases. cAMP is the downstream mediator of G proteins which activate adenylate cyclase upon ligand binding. Many different drugs, neurotransmitters and hormones exert their cellular effects by modulating adenylate cyclase activity and thus raising or lowering intracellular cAMP concentrations.
