For the most part, one of the differences between the Luminex 100 and the Luminex 200 machines is that a thumbwheel has been installed for probe height adjustment in the 200 versus a hex screw in the 100.

The doublet discriminator gate is automatically set at 8,000-13,500 and will exclude doublets or other larger bead aggregates.

The CAL1 setting sets the voltage of the photomultiplier tube (PMT) for the laser that detects bead colors and defines the bead number.

The CAL2 setting sets the voltage of the photomultiplier tube (PMT) for the laser that detects the reporter.

Experiments using Beadlyte® cytokine detection systems have shown that the high calibration does not increase sensitivity. Although the MFI values are higher, the background also increases.

Reading a plate using “high” RP1 targets may provide increased detection in some assays. If using the higher calibration settings, we recommend that the assay first be read based on the low target value, then re-read based on the high target value. This will prevent the loss of data that may be in the high range of the assay.
