Product Overview:
Dual Detect CELISA is an efficient Cell-based ELISA (CELISA) assay that eliminates the need to prepare cell lysates. It enables for the measurement of the relative levels of both the total and the phosphorylated forms of the target in whole cells in a convenient 96 well microplate format using dual fluorescent detection. This allows for the normalization of total protein between wells, thus allowing a more accurate measurement of phosphorylation levels among cell and/or growth condition differentials.
Cells are cultured directly in the microplate and stimulated as desired. They are then fixed, and then simultaneously incubated with two primary antibodies: One is a phosphospecific antibody directed against the phosphorylated epitope of the protein that correlates with it activation and the other is a total antibody that recognizes the target protein, regardless of phosphorylation status. The levels of both the total target protein and the phosphorylated protein are measured simultaneously in a single well using a double immunoenzymatic labeling procedure that utilize two spectrally distinct fluorogenic substrates for HRP and AP. The fluorescence of the phosphorylated protein is normalized to the total protein in each well thus reducing well-to-well variations. The ATF2 (Thr69/71) kit contains the components required to run the Dual Detect CELISA using fluorogenic substrates to measure both total ATF2 and phosphorylated ATF2 (Thr69/71) in the context of whole cells.

Background Information:
Activating transcription factor 2 (ATF2) is a member of the basic region leucine zipper (bZIP) family of proteins. Transcriptionally active ATF2 recognizes and binds to specific ATF/CRE motifs as homo- or heterodimers with other bZIP transcription factors of the ATF/CREB or AP-1 families. ATF2 is regulated by phosphorylation on Thr69/71. Two major kinases known to phosphorylate these sites are the two Stress Activated Protein Kinases (SAPK) Jun and p38 that are mitogen activated protein kinases (MAPKs). Phosphorylation and dimerization are required for the full transcriptional activity of ATF2. Transcriptional regulation by ATF2 has been implicated in mediating many diverse cellular processes such as growth, differentiation, immune and stress responses, inflammatory cytokine signaling, and oncogenic transformation.
